dc.description.abstract | Considerando che gli oligodendrocyte progenitor cells (OPCs) generano gli oligodendrociti maturi e che quindi sono coinvolte nel processo di rimielinizzazione si può ipotizzare che nerve glial antigen 2 (NG2), marker delle OPCs, possa avere un ruolo nella patogenesi autoimmune della sclerosi multipla (SM), soprattutto che possa essere bersaglio della risposta auto-anticorpale determinata dall’attivazione dei linfociti B.
Per investigare il ruolo antigenico di NG2, abbiamo ricercato ed identificato in campioni di liquor di pazienti SM auto-anticorpi anti-NG2 (α-NG2) attraverso esperimenti ELISA. Inoltre abbiamo correlato la presenza di α-NG2 in liquor di pazienti SM con lo stato clinico dei pazienti SM identificando una sotto-popolazione di pazienti SM che presenta attività auto-anticorpale e che risulta avere una patologia più grave. Suggeriamo quindi che gli α-NG2 possano essere considerati come un biomarcatore prognostico di tipo negativo.
Successivamente abbiamo esaminato quale fosse l’epitopo di NG2 riconosciuto dall’auto-Ab. Questo consentirebbe di valutare se gli α-NG2 possano interferire in qualche maniera con una data funzione relativa al dominio riconosciuto. Grazie ad esperimenti in dot plot è emerso che il pattern tipico dei pazienti SM positivi è la contemporanea positività ai pool A e C. In particolare è tipica dei soli pazienti SM positivi la positività al pool A, ossia la porzione N-terminale di NG2 presentante i laminin G-like domain coinvolti nell’interazione con la matrice extracellulare.
Infine, grazie ad esperimento in vitro di immunofluorescenza, abbiamo dimostrato che gli α-NG2 presenti nei pazienti SM positivi sembrano riconoscere in maniera specifica le OPCs.
Alla luce di quanto descritto, abbiamo determinato la presenza di α-NG2 in pazienti affetti da SM e riteniamo che tali Ab possano influenzare l’attività delle OPCs e potrebbero costituire un possibile biomarcatore prognostico negativo in pazienti con SM. | it_IT |
dc.description.abstract | The nerve-glial antigen 2 (NG2)-espressing oligodendrocyte progenitor cells (OPCs) generate mature myelinating oligodendrocytes and are therefore involved in the remyelination process. We hypothesised that NG2 may play a role in the autoimmune pathogenesis of multiple sclerosis (MS), especially that it may be a target of the auto-antibody response mediated by B lymphocyte activation.
In order to understand whether or not NG2 may play a role of auto-antigen, we investigated the presence of auto-antibodies anti-NG2 (α-NG2) in CSF samples from MS patients by ELISA. We determine their presence in 34,66% of MS patients and those patients displayed a more aggressive disease course. Accordingly, we suggest that α-NG2 may be considered as a negative prognostic biomarker.
Then we examined which NG2 epitope was recognised by the auto-Ab. This would allow us to assess whether α-NG2 could interfere with a given function of NG2. Dot plot experiments revelaed that the α-NG2 expressed by MS patients recognized the NG2 N-terminal where is located the laminin G-like domains which are involved in the interaction with the extracellular matrix.
Finally, through in vitro immunofluorescence experiments, we demonstrated that α-NG2 in MS positive patients seem to recognise OPCs.
In conclusion, we determined the presence of α-NG2 in MS patients and we suggest that these Ab may influence the activity of OPCs and could be a possible negative prognostic biomarker in MS patients. | en_UK |
