dc.description.abstractLa Tripanosomiasi Africana Umana e la Leishmaniosi sono infezioni protozarie causate rispettivamente dai parassiti Tripanosoma brucei e Leishmania. Sono annoverate tra le malattie tropicali neglette (NTDs), patologie per le quali le cure sono limitate in termini di disponibilità, sicurezza ed efficacia. L’emergere di parassiti resistenti ai farmaci inoltre pregiudica l’esito positivo della terapia. In questo contesto durante il mio lavoro di tesi ho sintetizzato e saggiato in vitro una nuova serie di molecole a struttura guanidino benzimidazolica per l’attività antiprotozoaria attribuibile all’inibizione della Diidrofolato reduttasi (DHFR) dei parassiti. Alcuni composti hanno esibito una promettente attività nel basso range micromolare, ma non hanno confermato l’ipotesi di inibizione competitiva della DHFR, suggerendo l’interesse ad approfondire il meccanismo d’azione di questi composti in studi successivi.it_IT
dc.description.abstractHuman African Trypanosomiasis and Leishmaniasis are protozoan infections caused by Trypanosoma brucei and Leishmania, respectively. They are included among the Neglected Tropical Diseases (NTDs), for which treatments show several limitations in terms of availability, safety and efficacy. Moreover, the emergence of drug-resistant parasites jeopardizes the positive outcome of the therapy. In this context, during my thesis work, I have synthesized a novel series of molecules based on the guanidino benzimidazole scaffold, that I have tested in vitro for the antiprotozoan activity, attributable to a competitive inhibition of Dihydrofolate reductase (DHFR). Some compounds have exhibited a promising activity in the low micromolar range, but they have not confirmed the competitive inhibition hypothesis of DHFR, suggesting the interest to deepen their mechanism of action in a further step.en_UK
