dc.description.abstractLe numerose interazioni con diversi pathways fisiologici e patologici hanno reso le p38 MAPK chinasi un interessante target, potenzialmente utile nel trattamento di disturbi infiammatori, tumori e malattie neurodegenerative, quali Alzheimer e Parkinson. Scopo della presente tesi è stata la sintesi e la valutazione biologica di composti a struttura imidazo-pirazolica, portanti in posizione 6 diversi sostituenti ammidici, strutturalmente molto simili ad altri derivati che, già in passato, erano risultati efficaci nell’inibire la fosforilazione di p38MAPK in cellule HUVEC. Questa nuova serie di composti è stata testata su piastrine umane, un modello biologico veloce e a basso costo, che ci ha permesso di valutare sia l’inibizione della fosforilazione di p38MAPK, che l’inibizione della produzione di radicali liberi (ROS). I composti sintetizzati hanno mostrato una buona attività, poiché sono risultati in grado di inibire la fosforilazione di p38 e la produzione di ROS, con IC50 comprese tra 80 e 100 μM.it_IT
dc.description.abstractThe numerous interactions of p38MAPK with different physiological and pathological pathways have made it an interesting target, potentially useful for the treatment of infiammatory diseases, cancer and neurodegenerative disorders, such as Alzheimer and Parkinson disease. The purpose of the following thesis has been the synthesis and the biological evaluation of imidazo-pyrazole compounds, carrying in position 6 various amide substituents, highly similar to the structure of other derivatives, that in the past was shown to be effective in the inhibition of p38MAPK phosphorylation in HUVEC cells. This new series of compounds have been tested on human platelets, a fast and cheap biological model, that has allowed us to evaluate both the p38MAPK phosphorilation and the reactive oxygen species (ROS) production inhibition. The synthesized compounds have shown a promising activity, because they have been found able to inhibit p38 phosphorilation, and ROS production, with IC50 between 80 and 100 μM.en_UK
