dc.description.abstract | Introduzione e Scopo dello Studio
Scopo dello studio è valutare la macro- e micro-struttura di 5 differenti impianti, valutandone la topografia di superficie e i valori di rugosità, i quali, insieme alla chimica superficiale, anch’essa oggetto di analisi, concorrono a garantire un’adeguata osteointegrazione. La seconda fase della ricerca ha lo scopo di valutare l’eventuale risposta infiammatoria indotta in vitro nei macrofagi a contatto con gli impianti precedentemente analizzati.
Metodi
Sono stati analizzati 1 impianto in zirconia (NobelPearl di Nobel Biocare) e 4 impianti in titanio: Syra (Sweden&Martina), Prama (Sweden&Martina), T3 (Biomet3i) e Shard (Mech&Human), tramite analisi SEM-EDX e valutazione quantitativa della rugosità superficiale (1 campione), analisi chimica XPS (1 campione) e analisi dell’espressione genica su cellule macrofagiche murine J774a.1 (3 campioni). Per quest’ultima fase, dopo 4h di contatto dei macrofagi con gli impianti, è stato estratto l’RNA totale, retrotrascritto a cDNA ed è stata valutata l’espressione genica dei macrofagi mediante PCR quantitativa (qPCR) in relazione ai geni GAPDH, YWHAZ, IL1β, IL6, TNFα, NOS2, MMP9, MMP8, TIMP3. I risultati di questa analisi sono stati analizzati statisticamente e comparati con il controllo positivo e negativo.
Risultati
I valori di rugosità rientrano nei range considerati “ideali” secondo la letteratura, così come i valori di pulizia chimica, nei quali solo 3iT3 risulta avere un lieve accumulo di C e Si. La valutazione della risposta infiammatoria ha dimostrato che nessun impianto ha indotto una risposta infiammatoria significativa nei macrofagi, anche se 3iT3 ha presentato un leggero effetto pro-infiammatorio rispetto agli altri campioni.
Conclusioni
Tutti gli impianti esaminati risultano ben concepiti in termini di topografia e rugosità superficiale, puliti in termini di residui di tipo chimico e non inducono stimoli infiammatori significativi nelle cellule macrofagiche. | it_IT |
dc.description.abstract | Intro and Purpose of the Study
The aim of this study is to make an evaluation of the macro- and micro-structure of 5 different implants, analyzing their surface topography and roughness values. These two parameters, together with surface chemistry, which was also studied in this analysis, are important to ensure adequate osseointegration.
The second phase of the research aims to evaluate the possible inflammatory response induced in vitro in macrophages in contact with the previously analyzed implants.
Methods
1 zirconia implant (NobelPearl of Nobel Biocare) and 4 titanium implants, Syra (Sweden&Martina), Prama (Sweden&Martina), T3 (Biomet3i) and Shard (Mech&Human), were analyzed through SEM-EDX analysis and quantitative evaluation of surface roughness (1 sample), XPS chemical analysis (1 sample) and analysis of gene expression on murine macrophagic cells J774a.1 (3 samples). For this last phase, after 4h of contact of the macrophages with the implants, the total RNA was extracted, retrotranscribed to cDNA and the gene expression of the macrophages was evaluated by quantitative PCR (qPCR) in relation to the following genes: GAPDH, YWHAZ, IL1β, IL6, TNFα, NOS2, MMP9, MMP8, TIMP3. The results were statistically analyzed and compared with positive and negative controls.
Results
The roughness values recorded fall within the ranges considered "ideal" according to the literature, as well as the chemical cleaning values, in which only 3iT3 showed a slight accumulation of C and Si. The evaluation of the inflammatory response has shown that no implant triggered a significant inflammatory response in macrophages, although 3iT3 exhibited a slight pro-inflammatory effect compared to other samples.
Conclusions
All the samples were well-conceived in terms of topography and surface roughness, clean in terms of chemical residues, and without any inflammatory stimulus on the examined macrophagic cells. | en_UK |
