| it_IT |
| dc.description.abstract | The skull bone marrow (SkBM) and its compartments at the interface with the central nervous system (CNS) are emerging as increasingly relevant players in the inflammatory processes underlying autoimmune neuroinflammatory diseases. This project aims to design synthetic promoters to be inserted into lentiviral vectors for tracing haematopoietic stem and progenitor cells (HSPCs), with particular focus on the populations residing in the SkBM. The vectors will be delivered in vivo in reporter mouse models and, in the future, in models of experimental autoimmune encephalomyelitis (EAE), in order to study the kinetics of the traced populations and their contribution to inflammation.
The work is organised in three stages. The first is an in silico analysis, in which the integration of RNA-seq and ATAC-seq data allowed the selection of several transcription factor binding sites (TFBSs). The second is experimental validation: ATAC-seq libraries were generated from ex vivo samples following the protocol of Buenrostro et al. (1). The third consists of generating a preliminary construct carrying the elements shared with the future HSPC-specific vector (EGFP-Cre), placed however under the control of the constitutive CMV promoter, which will be replaced by the specific promoter once defined.
The in silico analyses identified several TFBSs, including Klf10 and Plagl2, shared by both, while the ex vivo ATAC-seq libraries identified further candidates, notably Gata2 and Gata3, both known for their role in the emergence and maintenance of haematopoietic stem cells. These results provide the basis for the design and validation of the synthetic promoters. In vivo testing of the EGFP-Cre construct allowed the concentration and volume of the viral vector for intracalvarial delivery to be optimised. It also revealed a stronger signal in the dura mater, the significance of which will need to be clarified by further experiments. | en_UK |
