Functional maturation of hiPSC-derived neuronal networks on microelectrode arrays (MEAs) is usually characterized with spike-based metrics, while the low-frequency local field potential (LFP), which reflects subthreshold population activity and shares its origin with the EEG, is typically filtered out. Whether LFP maturation signatures emerge in two-dimensional (2D) cortical cultures remains largely unexplored. This thesis examined the longitudinal evolution of the LFP in 2D cortical networks derived from the TUBA1B hiPSC line, recorded at four timepoints between DIV 47 and 59. Two batches recorded on systems with different low-frequency limits were compared to exclude technical bias; since the older system attenuated slow components, spectral analyses used the broadband LFP, and spike metrics were reported for both. Power distribution across canonical bands, the aperiodic component, and delta-broadband phase-amplitude coupling (PAC) were evaluated and combined by principal component analysis into a maturation index, the LFP-based neural activity score (LFP-NAS). Spike metrics followed expected developmental trajectories. Spectral power shifted from low to high frequencies with a stable aperiodic component, indicating band-specific changes rather than global reorganisation. Delta-broadband PAC emerged and strengthened over time, consistent with increasing population-level rhythm locking. The LFP-NAS tracked single-well maturation, rising significantly with DIV from negative to positive values, driven mainly by gamma power, the two PAC measures, relative delta power, and the delta-to-gamma ratio. These results show that the LFP in 2D hiPSC-derived networks is an informative readout of functional maturation, and that the LFP-NAS complements spike-based characterisation, with applications in disease modelling and drug screening.en_UK
