Mutations of the human X-linked gene CLCN4 cause neurological phenotypes in human patients. CLCN4 encodes the membrane transporter protein in endosomes, ClC-4, which functions as a 2Cl^- / H^+ antiporter. Studying its mutations and comparing its functionality via electrophysiological assays could help understand its role, still not fully clear, in physiological as well as pathological scenarios.
In this work the variant Y177N, identified in a patient who shows severe delays in motor and communication development, has been studied. Y177N is so far an undescribed de novo variant.
The mutation has been reproduced in vitro and the functionality of the corresponding antiporter has been analyzed by two-electrode voltage-clamp after exogenous expression in Xenopus laevis oocytes. The mutated protein has also been expressed in HeLa human cells, where its interaction with TMEM9B, an inhibiting protein of ClC-4, has been studied. This resulted in the classification of Y177N as a gain of function variant, contributing to the knowledge regarding ClC-4 and CLCN4-related diseases, and joining the continuous work of researchers who collaborate with the association Cure CLCN-4.
